The research

PhD thesis: Bicyclo[1.1.1]pentane Analogues of Quinolone Antibiotics, supervised by Dr Marc Kimber at Loughborough University.

1

The problem

Quinolones are among the most widely used antibiotic classes, but resistance to them keeps rising. Antimicrobial resistance is one of the biggest threats in modern medicine, and new chemical starting points are needed.

2

The idea

Many drugs are built from flat aromatic rings. Bicyclo[1.1.1]pentane (BCP) is a compact, rigid cage that can stand in for those flat pieces as a bioisostere, adding three dimensional shape that can change how a molecule behaves in the body.

3

The chemistry

Strained cages are hard to attach to drug scaffolds. Dolapo develops and optimises multistep synthetic routes to make these analogues, then studies how changes to their structure affect their properties.
